چکیده

Ethidium bromide (EB), acridine orange (AO), methylene blue (MB) and other fluorescent compounds are often used to probe DNA structure in drug–DNA and protein–DNA interactions. They bind nucleic acids via intercalative mode and cause major changes to DNA and RNA structures. The aim of this study was to examine the stability and structural features of calf-thymus DNA complexes with EB, AO andMB in aqueous solution, using constant DNA concentration (12.5 mM) and various pigment/DNA(P) ratios of 1/40, 1/20, 1/ 10, 1/4 and 1/2. FTIR, UV–visible spectroscopy and isothermal titration calorimetry (ITC) are used to determine the ligand intercalation and external binding modes, the binding constant and the stability of pigment–DNA complexes in aqueous solution. Structural analysis showedmajor intercalation of EB,AO andMB into polynucleotidesG–C andA–T base pairs withminor external binding and overall binding constants ofKEB = 6.58 · 10 M ,KAO = 2.69 · 10 M 1 andKMB = 2.13 ·10 M . The overall binding constants estimated byUV– visible spectroscopy are consistent with thermodynamic data obtained by ITC, which showed DH(EB) = 13.58 kJ/mol, DH(AO) = 14.63 kJ/mol andDH(MB) = 13.87 kJ/mol with dissociation constants ofKEB = 15 lM,KAO = 36 lMandKMB = 46 lM. 2006 Elsevier B.V. Study of Peculiarities of Differential Absorption Spectra of Complexes of Acridine Orange, Ethidium Bromide and Methylene Blue with Dna

Interaction of intercalators: ethidium bromide (EtBr), methylene blue (MB) and acridine orange (AO), with DNA at 0.02 M ionic strength of the solution has been studied by differential spectroscopy method. It was revealed that EtBr binds to DNA by intercalation, semi-intercalation and electrostatic modes; MB by semiintercalation and electrostatic modes and AO by intercalation and electrostatic m...

Changes in superhelical density of closed circular deoxyribonucleic acid by intercalation of anti-R-plasmid drugs and primaquine.

The following compounds, which possess anti-R-plasmid activity (Hahn and Ciak, 1976) in decreasing order, were shown by viscometric titration to change systematically the superhelical density of closed circular PM2 deoxyribonucleic acid in the manner of intercalators: ethidium bromide, quinacrine, acridine orange, quinine, chlorpromazine, chloroquine, and methylene blue. The same effect was cau...

Melting Parameters of Dna Complexes with Several Intercalators

Comparative study of DNA complexes with acridine orange (AO), ethidium bromide (EtBr) and methylene blue (MB) has been carried out at 0.02 M ionic strength of solution in 0 < r  0.33, r = [ligand] / [DNA] range. The values of changes of melting parameters ((1/Tm) and (T/Tm)) of these complexes were determined. From the obtained data it was revealed that the dependence of (1/Tm) enhances in...

Effects of Aflatoxin B1 and Fumonisin B1 on the Viability and Induction of Apoptosis in Rat Primary Hepatocytes

The present study evaluated the effect of aflatoxin B(1) (AFB(1)) and fumonisin B(1) (FB(1)) either alone, or in association, on rat primary hepatocyte cultures. Cell viability was assessed by flow cytometry after propidium iodine intercalation. DNA fragmentation and apoptosis were assessed by agarose gel electrophoresis and acridine orange and ethidium bromide staining. At the concentrations o...
